Research Overview

NAD+ (β-Nicotinamide Adenine Dinucleotide) is a dinucleotide coenzyme found in all living cells, where it serves as the primary electron carrier in oxidative phosphorylation. Its oxidised form (NAD+) accepts electrons from metabolic substrates, becoming NADH, which then donates electrons to the mitochondrial electron transport chain to generate ATP.

Beyond its classical role in bioenergetics, NAD+ has emerged as a critical signalling molecule. It is the obligate substrate for sirtuins (SIRT1–7), a family of NAD+-dependent deacylases that regulate gene expression, mitochondrial biogenesis, inflammation, and stress responses. NAD+ also fuels PARP enzymes (poly-ADP-ribose polymerases), which are central to DNA damage detection and repair. Additionally, it is consumed by CD38, a major NAD+-consuming enzyme whose activity increases with age.
